1 Introduction
Histamine is a potentially harmful nitrogen-containing low molecular weight organic compound. Eating certain fish or other animals containing a certain amount of histamine can cause allergic food poisoning. Histamine poisoning is characterized by pain, light symptoms, quick recovery, short incubation period, and is manifested as mental weakness, allergies, skin flushing, vomiting, diarrhea, etc.

GB2733-2015 "Fresh and Frozen Animal Aquatic Products" stipulates: high histamine fish 40mg/100g, other seawater fish 20mg/100g.
3 Detection principle
After histamine is extracted, it reacts with the detection reagent to form a colored compound. The color depth is related to the histamine content, and the approximate content can be obtained from the color card ratio.

6 sample processing and determination
6.1 Take reagent F before experiment and pour it into reagent G, mix well as a reaction reagent.
6.2 Weigh 2g of pulverized fish in a 7mL centrifuge tube, add reagent A 3mL, shake vigorously for 5 minutes, let stand for 5 minutes and take supernatant as the liquid to be tested, filter or centrifuge if necessary.
6 Take 0.5mL of the solution to be tested in a 10mL centrifuge tube, add 1 drop of reagent B to make the liquid alkaline, then add 1mL of reagent C, shake vigorously for 5 minutes and then centrifuge at 4000r/min for 2 minutes, the supernatant is used as the solution to be tested.
6 Take 0.5mL of the supernatant in another 7mL centrifuge tube, add 1mL of reagent D, shake vigorously for 5 minutes, and let stand for stratification and take the lower layer as the solution to be tested.
6 Remove 0.5mL of the lower liquid in a 2mL centrifuge tube, add 1mL of reagent E and mix well, and then add 0.5mL of the mixture of reagent F and reagent G, and shake well. After standing for 5 minutes, compare with the color card, and the color scale with similar colors is the actual concentration of histamine in the sample.
7 Precautions
7.1 The experimental water is purified water or distilled water.
