Sulfonamide drug colloidal gold rapid detection card instruction manual
1 principle and use
This product is made of the principle of competitive inhibition colloidal gold immunochromatography, which is used for rapid detection of sulfonamide drug residues in aquatic products, livestock and poultry tissue samples (chicken, duck, fish, shrimp). After the sample solution is dripped into the sample hole of the detection card, the sulfonamide in the sample solution binds to the gold standard antibody, thereby preventing the gold standard antibody from binding to the sulfonamide conjugate on the cellulose membrane. When the sulfonamide content in the sample solution is greater than the detection limit, the detection line T does not show color or is significantly lighter than the C line, and the result is positive; when the sulfonamide content in the sample solution is less than the detection limit, the detection line shows purplish red (the T line is consistent with or deeper than the C line), and
2 Technical indicators
Total sulfonamide drug detection limit: 100 ppb (μg/kg)
3 You need to bring your own tools
Homogenizer, forceps, electronic balance (0.01-100 g), pipette (0.1-1 mL), vortex meter, low speed centrifuge, timer.
4 sample pretreatment
[tissue sample]
4 Take a certain amount of chopped aquatic products (fish and shrimp and other aquatic products) or livestock and poultry tissue samples, homogenize with a homogenizer;
4 Weigh about 0.25 grams of homogeneous material in the detection tube;
4 Add 1 mL of dilution and shake violently for 1 minute;
4 Centrifuge for 5 minutes at 4000 rpm at room temperature, supernatant as the liquid to be tested;
5 sample detection
5 Tear off the aluminum foil bag, take out the detection card, and place it horizontally on the table;
5.2 Pipette 120 µL (or take 4 drops with a dropper) of the liquid to be tested, and add it vertically to the sample hole (S) of the detection card;
5 Start timing after adding the sample, the results should be read in 8-10 minutes, and read at other times No.
6 the results judged
negative: the control line (C) appears purplish red line, the detection line (T) is darker or as deep as the C line, indicating that the residual concentration of sulfonamides in the sample is lower than the detection limit or no sulfonamides.
Positive: The control line (C) appears purplish red line, the detection line (T) does not show color or the T line shows color is significantly lighter than the C line, indicating that the residual concentration of sulfonamides in the sample is higher than the detection limit.
Failure: In the detection window, the control line (C) does not appear purplish-red line.
7 Precautions
7 Products that have expired or damaged aluminum foil bags cannot be used.
7 When the test card is taken out of the refrigerator, it should be restored to room temperature and opened. The opened test card should be used as soon as possible to avoid failure after moisture.
7 When using the centrifuge, it needs to be trimmed and placed symmetrically.
7 The straw is disposable and cannot be mixed to avoid cross-contamination.
7 The sample solution to be tested should be clear, no cloudy particles, and no bacterial contamination, otherwise it will easily lead to abnormal phenomena such as blockage and inconspicuous color development, which will affect the judgment of the experimental results.
7.6 If you need to directly test the standard product, you need to prepare the diluent in the kit.
8 Storage and shelf life
Storage conditions: 4-30 ° C Store in the dark, do not freeze.
Shelf life: valid period 1 year, see the box for the production date.
