1 Principle and Use

This product is made of the principle of competitive inhibition of colloidal gold immunochromatography for detecting diazepam drug residues in aquatic livestock and poultry tissues (fish, shrimp, chickens, ducks, pigs, etc.). The whole detection process only takes about 20 minutes.

After the sample solution is dropped into the sample hole of the detection card, the diazepam in the sample solution binds to the gold standard antibody, thereby preventing the gold standard antibody from binding to the diazepam conjugate on the cellulose membrane. When the content of diazepam in the sample solution is greater than the detection limit, the detection line does not show color, and the result is positive; when the content of diazepam in the sample solution is less than the detection limit, the detection line shows purplish red (the T line is the same as the C line or deeper), and the result is negative.

3 required equipment and reagents

Homogenizer/cooking machine, vortex mixer (optional), centrifuge, electronic balance, pipette (0.1-1 mL), timer

4 Sample pretreatment

Tissue samples should be stored refrigerated in the dark.

4 Take a certain amount of chopped adipose tissue samples and homogenize them with a homogenizer;

4 Weigh about 0.25g of homogeneous material in a centrifuge tube;

4 Add 0.75 mL of diazepam dedicated dilution and shake violently for 5 minutes;

4 Centrifuge for 5 minutes at room temperature (20-25 ° C) 4000 rpm;

4 Aspirate supernatant 120 µL (about 4 drops) to be inspected.

5 Sample Testing

5.1 Tear the test card aluminum foil packaging bag, remove the test card, and place it on a flat, clean countertop.

5 Absorb 120 µL of the sample solution to be tested in the gold standard micropores, repeatedly blow and mix (at least 10 times) until the red substance in the gold standard micropores is completely dissolved, wait 2 minutes;

5 All the liquid in the suction micropores is added dropwise to the detection card. Add the sample hole;

5 Start timing after adding the sample, the result should be judged in 8-10 minutes, and the other time interpretation is invalid. 6 Results Judgment

Negative: The T-line of the test line shows that it is deeper or as deep as the C-line of the quality control line, indicating that the content of diazepam in the sample is lower than the detection limit or there is no diazepam residue;

Positive: The T-line of the test line has no color, indicating that the concentration of diazepam in the sample is higher than the detection limit;

Invalid: In the detection window, the control line (C) does not appear purplish-red line.

7 Precautions

7 Products that have expired or damaged aluminum foil bags cannot be used.

7 When the test card is taken out of the refrigerator, it should be restored to room temperature and opened. The opened test card should be used as soon as possible to avoid failure after moisture.

7 Do not touch the white film surface in the center of the test card.

7 The sample solution to be tested should be clear, free of cloudy particles, and free of bacterial contamination, otherwise it will easily lead to abnormal phenomena such as blockage and insignificant color development, which will affect the judgment of experimental results.

8 Storage and shelf life

8 Storage conditions: The kit is stored in a dry environment at 4-30 ° C.
