Zearalenone Colloidal Gold Rapid Detection Card Instruction Manual
1 Principle and Use
Zearalenone The main toxin-producing bacteria are Fusarium. Zearalenone mainly contaminates corn, wheat, rice, barley, millet and oats and other grains. This product is made of the principle of competitive inhibition of colloidal gold immunochromatography, which is used to detect zearalenone residues in grains and feeds. After the sample solution is dropped into the sample hole of the detection card, the zearalenone in the sample solution binds to the gold standard antibody, thereby preventing the gold standard antibody from binding to the zearalenone conjugate on the cellulose membrane. The result is judged according to the color depth of the T-line and the C-line.
2 Technical indicators
Detection limit: 200 μg//kg
3 Need to bring your own tools
balance, pipette, 15 ml low speed centrifuge, timer.
4 sample pretreatment
4. 1 The grain or feed sample to be tested is crushed into granules (do not grind into powder), and 1 gram of the sample is weighed in a 15 mL centrifuge tube; 117277984004.2 Add 9 mL of 20% methanol water, mix violently for 1 minute, and centrifuge at 4000 rpm for 5 minutes. The supernatant is used as the liquid to be tested;
5 sample detection
5.1 Tear off the aluminum foil bag of the detection card, remove the detection card/gold microporous/dropper (a total of 3 kinds of consumables), and place it on a flat, clean countertop.
5 with pipette or aluminum foil bag matching dropper to absorb the above liquid to be tested 120 µL (about 4 drops) in the gold standard micropores, horizontal stand for 2 minutes, and then repeatedly blow the purple material in the micropores with a dropper until completely dissolved; all the liquid in the gold standard micropores is added dropwise to the sample well (S) of the detection card;
5 Start timing after adding the sample, and leave it at room temperature for 8-10 minutes to judge the result, and other time interpretation is invalid.
6 the result
negative: the control line (C) and the detection line (T) both appear purplish-red lines, indicating that the concentration of zearalenone in the sample is lower than the detection limit or no zearalenone residue.
Positive: Purple-red line appears on control line C, and the detection line (T) does not show color, indicating that the concentration of zearalenone in corn is higher than the detection limit.
Failure: Purple-red line does not appear on control line C.
7 Precautions
7 Products that have expired or damaged aluminum foil bags cannot be used.
7 When the test card is taken out of the refrigerator, it should be restored to room temperature and opened. The opened test card should be used as soon as possible to avoid moisture.
7 Do not touch the white film surface in the center of the test card.
7.4 The sample solution to be tested should be clear, free of cloudy particles, and free of bacterial pollution, otherwise it will easily lead to abnormal phenomena such as blockage and insignificant color development, which will affect the judgment of experimental results.
8 Storage and shelf life
8.1 Storage conditions: 4-30 ℃ Store in the dark, do not freeze.
8 Shelf life: 1 year, see the outer packaging for the production date.
