1 Principle and use

The amantadine detection card is used for rapid detection of amantadine residues in poultry egg samples. This product is made by applying the principle of competitive inhibition of colloidal gold immunochromatography and is used to detect amantadine in samples. After the sample solution is dropped into the sample well of the detection card, the amantadine in the sample solution binds to the gold standard antibody, thereby preventing the gold standard antibody from binding to the amantadine conjugate on the cellulose membrane. The result is judged according to the color development depth of the T-line and the C-line.

4 sample pretreatment

(1) Break the fresh eggs into the cup and mix well (egg white and egg yolk are well mixed);

(2) Absorb 0.2 grams of the well-mixed egg sample in the test tube, and then add 1.2 mL dilution;

(3) Shake violently for 1 minute, which is the liquid to be tested;

5 sample detection

(1) Tear open the aluminum foil bag of the test card, take out the test card/gold standard microporous/dropper (a total of 3 kinds of consumables), and place it on a flat and clean countertop.

(2) with a pipette or a dropper in an aluminum foil bag to absorb the above test solution 120 µL (about 4 drops) in the gold standard micropores, blow with a dropper until the purplish-red substance in the pores is completely dissolved, let stand horizontally, and wait for the reaction for 2 minutes; All liquids in the gold standard micropores are added dropwise to the sample well (S) of the detection card; (3) Start timing after adding the sample, and the result is judged after 8-10 minutes at room temperature. The result interpretation for more than 10 minutes is invalid.

6 the result judgment

negative (-): In the detection window, the purple line appears on the control line (C), and the color development of the detection line (T) is consistent with or deeper than that of the C line, indicating that the concentration of amantadine in the sample is lower than the detection limit or there is no amantadine.

Positive (+): In the detection window, the purplish-red line appears on the C line, and the T line does not develop color or is lighter than the C line, indicating that the amantadine concentration in the sample is higher than the detection limit.

invalid: In the detection window, the control line (C) does not appear purplish-red line.

7 Precautions

(1) Products that have expired or the aluminum foil bag is damaged cannot be used.

(2) When the test card is taken out of the refrigerator, it should be restored to room temperature and opened. The opened test card should be used as soon as possible to avoid failure after moisture.

(3) Do not touch the white film surface in the center of the test card.

(4) The liquid extraction dropper cannot be mixed to avoid cross-contamination.

(5) The liquid to be tested should be clear, free of cloudy particles, and free of bacterial contamination, otherwise it will easily lead to abnormal phenomena such as blockage and inconspicuous color development, which will affect the judgment of experimental results.

(6) Please store the reagent in a place that is not easy for children to contact, and do not eat the equipped reagent.

(7) If you need to directly test the standard product, please prepare it with the diluent provided in the kit.
