1 Principle and Use

Quinolyethanol Metabolite Detection Card is used for rapid detection of quinolyethanol metabolite residues in tissue samples. This product is made by applying the principle of competitive inhibition colloidal gold immunochromatography and is used to detect quinolyethanol in samples. After the sample solution is dropped into the sample well of the detection card, the quinolyethanol in the sample solution binds to the gold standard antibody, thereby preventing the gold standard antibody from binding to the quinolyethanol conjugate on the cellulose membrane. 4 sample pretreatment

Tissue samples should be kept refrigerated in the dark.

(1) Take a certain amount of chopped adipose tissue samples and homogenize them with a homogenizer;

(2) Weigh about 5 grams of homogeneous material in a 50 mL centrifuge tube;

(3) Add 6 mL of reagent A and shake violently for 3 minutes;

(4) Add 8 mL of ethyl acetate and gently invert it up and down for 2 minutes (do not shake vigorously, otherwise emulsification will occur resulting in less than 5 mL of sampling);

(5) Centrifuge at 4000 rpm for 5 minutes;

(6) Pipette 5 mL of supernatant in a 10 mL centrifuge tube and blow dry at 65 ° C with nitrogen (empty) air;

(7) Add 0.5 mL of quinoethanol metabolite complex solution to the blow-dried centrifuge tube and repeatedly pipette the liquid in the tube to fully dissolve the tube wall

5 sample testing

(1) Tear off the aluminum foil bag of the test card, take out the test card/gold standard microporous/dropper (a total of 3 kinds of consumables), and put it on a flat and clean table.

(2) with a pipette or a dropper in an aluminum foil bag to absorb the above test liquid 120 µL (about 4 drops) in the gold standard micropores, blow with a dropper until the purplish-red substance in the pores is completely dissolved, let stand horizontally, and wait for the reaction for 2 minutes; all the liquid in the gold standard micropores is added dropwise to the sample well (S) of the detection card;

(3) Start timing after adding the sample, and leave it at room temperature for 8-10 minutes to judge the result, and the other time is invalid.

negative (-): the purplish-red line appears on the control line (C), and the color development of the detection line (T) is consistent with or deeper than that of the C line, indicating that the concentration of the substance to be tested in the sample is lower than the detection limit or there is no

Positive (+): Purple red line appears on line C, T line does not develop color or is lighter than line C, indicating that the concentration of the substance to be tested in the sample is higher than the detection limit.

Invalid: Control line (C) does not appear purplish red line.

7 Precautions

(1) Products that have expired or the aluminum foil bag is damaged cannot be used.

(2) When the test card is taken out of the refrigerator, it should be restored to room temperature and opened. The opened test card should be used as soon as possible to avoid failure.

(3) Do not touch the white film surface in the center of the test card.

(4) The liquid dropper cannot be mixed to avoid cross-contamination.

(5) Please keep the reagent in a place that is not easy for children to contact. Do not eat the reagent equipped.

(6) If you need to directly test the standard product, please prepare it with the dilution provided in the kit.

8 Storage and shelf life
