1 Principle and Use

Chloramphenicol Detection Card is used for rapid detection of chloramphenicol (CAP) residues in poultry egg samples. This product is made using the principle of competitive inhibition colloidal gold immunochromatography for the detection of chloramphenicol in samples. After the sample solution is dropped into the sample hole of the detection card, the chloramphenicol in the sample solution binds to the gold standard antibody, thereby preventing the gold standard antibody from binding to the chloramphenicol conjugate on the cellulose membrane. The result is judged according to the color development depth of the T-line and the C-line.

4 sample pretreatment


1. Dilution method

(1) Break fresh eggs into a cup and mix well (egg white and egg yolk are well mixed);

(2) Absorb 0.2 grams (about 4 drops) of mixed egg sample in the detection tube, and then add 1.2 mL of dilution;

(3) Vibrate violently for 1 minute or repeatedly suck the liquid in the detection tube with a dropper (not less than 10 times), which is the liquid to be tested;

Second, nitrogen blowing method

(1) Break fresh eggs into a cup and mix well (egg white and egg yolk are well mixed).

(2) Weigh 2.000.05 g of homogenized fresh egg sample into a 15 mL centrifuge tube, add 0.5 mL of NaCl solution, vortex for 2 minutes, then add 3 mL of sample extractant, vortex for 2 minutes, 3000-4000 rpm Centrifuge for 5 minutes;

(3) Take 2 mL of upper liquid into a new 10 mL centrifuge tube and blow dry under a nitrogen stream or air flow in a water bath at 50-60 ° C. (4) Add 300 μL of sample dilution and vortex for 30 seconds for the solution to be tested.

5 sample detection

(1) Tear open the aluminum foil bag of the detection card, take out the detection card/gold standard micropore/dropper (a total of 3 kinds of consumables), and put it on a flat and clean countertop.

(2) Use a pipette or a dropper in the aluminum foil bag to absorb the above test liquid 120 µL (about 4 drops) in the gold standard micropore, blow with the dropper until the purplish-red substance in the hole is completely dissolved, let it stand horizontally, and wait for the reaction for 2 minutes; all the liquids in the gold standard micropore are added dropwise to the sample hole (S) of the detection card;

(3) Start timing after adding the sample, and leave it at room temperature for 8-10 minutes to judge the result. The interpretation is invalid at other times.

6 result judgment

Negative (-): The color development of the T-line is darker than that of the C-line or there is no significant difference from that of the C-line, indicating that there is no substance to be tested in the sample or its concentration is below the detection limit.

Positive (+): The color development of the T-line is significantly weaker than that of the C-line or the T-line does not develop color, indicating that the concentration of the substance to be tested in the sample is equal to or higher than the detection limit.

Invalid: No C-line appears, indicating that the operation process is incorrect or the test card has failed. In this case, the instructions should be read carefully again and re-tested with a new test card.

(5) The liquid to be tested should be clear, no cloudy particles, and no bacterial contamination, otherwise it will easily lead to abnormal phenomena such as blockage and inobvious color development, thus affecting the judgment of experimental results.

(6) Please store the reagent in a place that is not easy for children to contact, and do not eat the reagent equipped.

(7) If you need to directly test the standard, please prepare it with the dilution provided in the kit.
